Glutamine synthetase is an enzyme that plays an important role in nitrogen metabolism by catalyzing the condensation of glutamate and ammonia to form glutamine. In vitro: Previous studies found that DL- and L-alpha-aminoadipic acid (alpha-AA) have specific gliotoxin in vitro. HPLC analysis of cultures incubated with D- or L-α-AA and DL-[14C]-α-AA autoradiograms in the presence of D- or L-α-AA demonstrates stereospecificity of astroglial L-α-AA uptake. Both alpha-AA uptake by astrocytes and alpha-AA-induced glial cytotoxicity are sodium-dependent. Another study found that the L-isomer of α-aminoadipic acid was able to competitively inhibit the transporter, whereas the D-isomer of α-aminoadipic acid was ineffective. Furthermore, L-α aminoadipic acid was found to be a competitive inhibitor of glutamine synthetase and γ-glutamylcysteine synthetase. In contrast, the D-isomer of α-aminoadipic acid inhibits both enzymes much less strongly. In vivo: Animal studies have shown that La-aminoadipic acid can dose-dependently reduce levels of endogenous extracellular kynurenic acid in the hippocampus (endent fashion), although La-aminoadipic acid appears to be less effective than it does in reducing de novo kynurenic acid.
