In this model for assessing the effects of uricosuric drugs, probenecid reduced the urinary excretion of pyrazinecarboxylic acid, indicating an inverse relationship between the urinary excretion of urate and pyrazinecarboxylic acid. In in vitro studies, treatment of vesicles in the brush border membrane of the rat kidney with pyrazinecarboxylic acid increases urate uptake, which is inhibited by uricosuric drugs. PZA (pyrazinamide) is converted to pyrazinecarboxylic acid (POA) in plant cells and inhibits the activity of 1-aminocyclopropane-1-carboxylic acid oxidase (ACO), which catalyzes the final step in ethylene formation.
