Related Catalog Research Areas >> Others In Vitro EIT (S-ethylisothiourea) Hydrobromide is a selective inhibitor of type II NOS. EIT produced a dose-dependent and >95% inhibition of LPS-induced increases in plasma [NOx]. The ED50 value of EIT is 0.4 mg/kg. Pretreatment with L-arginine (but not D-arginine) prevented mortality without affecting type II NOS-dependent NO production, suggesting that toxicity may be due to inhibition of one of the other NOS isoforms (endothelial or neuronal) [1]. Selective NOS II antagonists attenuate but do not block shear stress-induced vasodilation in fetal lungs. Stimulation of NOS II activity, possibly from smooth muscle cells, partially contributes to the NO-mediated decrease in PVR (pulmonary vascular resistance) during shear stress-induced pulmonary vasodilation. Splenocytes stimulated with alloantigens in the presence of AMT or another selective iNOS inhibitor, S-ethylisothiourea (EIT), produced more interleukin (IL)-4 and IL-10 than cells stimulated in the absence of the iNOS inhibitor. Production of the Th1 cytokines IL-2 and interferon (IFN)-γ was not enhanced by inhibition of NO synthesis. Acute administration of EIT (380 nmol/h), another selective inhibitor of inducible nitric oxide synthase, also attenuated the pregnancy-induced increases in glomerular filtration rate and effective renal plasma flow.
